preparation | 1-tert-butoxycarbonylamide cyclohexanecarboxylic acid was prepared as follows: A round-bottom flask was charged with sodium hydroxide solution (43ml, 4mol/L), THF(40ml), and 1-aminocyclohexanoic acid (5.0g,34.96mmol), di-tert-butyl dicarbonate (9.13g,41.96mmol) was added to the reaction flask. After completion of the dropwise addition, the reaction system was stirred at room temperature overnight. Water (100ml) was added to the reaction system, acidified with hydrochloric acid (50ml, 1mol/L), then extracted with ethyl acetate (50ml × 2), and the organic phases were combined, the organic phase was washed with saturated brine (100ml × 2) and dried over anhydrous sodium sulfate. The crude product was purified by silica gel column chromatography (petroleum ether/ethyl acetate (V/V)= 4/1), the title compound 1-t-butoxycarbonylamide cyclohexanecarboxylic acid was obtained as a white solid (7.3g,86%). |
